Abstract:Using total DNA of Wild abortion (WA) type CMS line Zhenshan 97 A and its maintainer Zhenshan 97 B as template, a 1600 bp band was amplified only in Zhenshan 97 A by OPA12 which was screened out from one hundred primers. The 1600 bp band was also found in WA type Longtepu A, Dwarf abortion (DA) type Xieqingzao A, F 1 and F 2 individual plants of Zhenshan 97A/Minghui 63, but not found in maintainer Longtepu B and Xieqingzao B, using total DNA as template and OPA12 as primer. The results indicate that the 1600 bp band amplified by OPA12 is a specific band of the male sterile cytoplasms. Using mitochondrial DNA (mtDNA) as template and amplified by OPA12, the 1600 bp band was found in the 3 CMS lines, but not found in the 3 maintainers mentioned above. In addition to the 1600 bp band, two other bands (1000 bp and 700 bp) were found in DA type Xieqingzao A. These results indicated that the 1600 bp band derives from CMS mtDNA and can be used as a RAPD marker to distinguish A and B plants at seedling stage, and the bands other than the 1600 bp can be used to distinguish WA and DA cytoplasms.
