脱钙处理对鱼鳞蛋白酶解效果及其持钙能力的影响

摘    要:研究了盐酸脱钙对鱼鳞蛋白酶解效果及其酶解产物持钙活性的影响。结果表明,浓度0.4 mol/L的HCl溶液,鱼鳞粉与盐酸的料液比为1∶30,脱钙时间30 min可显著提高鱼鳞蛋白溶出率与胶原蛋白溶出率,脱钙率达87.90%;采用胰蛋白酶、胃蛋白酶与风味蛋白酶分步酶解,酶解产物的持钙能力达13.93 mg(钙)/g(蛋白),是胰蛋白酶酶解产物持钙能力的5.32倍(p<0.01)。当酶解产物蛋白质量浓度为2 mg/mL时,酶解产物与钙离子螯合的最佳条件为:螯合反应温度50℃,pH值6.0,蛋白与CaCl2质量比为6∶1,螯合反应时间40 min。

Effects of Demineralization on Protein Hydrolysis and Calcium Binding Abilities of Tilapia Fish Scales
Abstract:The influence of demineralization on protein hydrolysis and the calcium binding activity of fish scale are investigated.The results indicate that 0.4 mol/L HCl,ratio of fish scale powder to HCl solution 1∶30 and the decalcification time 30 min can help protein and collagen in fish scale to dissolve,and the decalcification rate is 87.90%.The calcium binding activity of protein hydrolysates sequentially digested by trypsin,pepsin,flavorzyme is 13.93 mg calcium/g protein,5.32 times higher than that of hydrolysates digested by trypsin.The optimum conditions to get the higher concentration of protein-calcium chelates are as follows: temperature 50 ℃,pH value 6.0,the mass ratio of protein to calcium chloride 6∶1,and binding time 40 min.
