枯草杆菌B_s501抗稻瘟病菌条件优化

摘    要:目的]提高枯草芽孢杆菌的拮抗作用。方法]通过正交试验和单因素试验研究了枯草杆菌Bs501的发酵基础培养基和最适发酵条件,明确了几丁质的添加量及添加时机,分析了二价金属离子对枯草杆菌发酵液的影响。结果]枯草杆菌在LB培养基上培养12 h达到对数生长末期,随后生长曲线快速下降,56 h出现小波峰。各因子对枯草杆菌Bs501生长量的影响为蛋白胨>蔗糖>酵母膏>K2HPO4,它们对枯草杆菌Bs501的抑菌活性为蔗糖>蛋白胨>酵母膏>K2HPO4。最佳基础培养基确定为3%蔗糖、1%蛋白胨、0.5%酵母膏0、.45%K2HPO4。最适发酵条件为:起始pH值7.0、培养温度30℃、发酵时间48 h。培养12 h添加0.5%几丁质诱导枯草芽孢杆菌Bs501产生拮抗物质的效果最显著。Ca2+可显著提高发酵液的抑菌活性。结论]该研究为枯草芽孢杆菌的工业深层发酵提供了技术参考。

Abstract:The aim of the research was to increase the antagonistic action of Bacillus subtilis.In orthogonal test and single-factor test,the basal fermentation medium and the optimum fermentation conditions for B.subtilis Bs501 were studied.The adding amount of chitin and its adding time were confirmed and the effects of divalent metal ions on the fermentation liquid of B.subtilis were analyzed.B.subtilis reached logarithm growth last stage after it was cultured on LB medium for 12 h and then its growth curve rapidly decreased and appeared a small peak at the 56th hour.The effects of all factors on the growth increment of B.subtilis Bs501 was peptone>sucrose>yeast extract > K2HPO4.Their bacteriostatic activities to B.subtilis Bs501 were as follows:sucrose>peptone>yeast extract>K2HPO4.The optimum basic medium was confirmed as follows: 3% sucrose,1% peptone,0.5% yeast extract and 0.45% K2HPO4.The optimum fermentation conditions were: initial pH value of 7.0,culture temperature of 30 ℃,fermentation time of 48 h.Adding 0.5% chitin to induce B.subtilis Bs501 for producing antagonistic substance after culturing for 12 h got most significant effect.Ca2+ could increase the bacteriostatic activity of fermentation liquid significantly.The research provides technical references for the industrial submerged fermentation of B.subtilis.
