玉龙雪山不同海拔急尖长苞冷杉遗传多样性分析

摘    要:利用4对SSR引物对玉龙雪山内5个海拔梯度的急尖长苞冷杉居群进行遗传多样性和遗传结构的研究。在物种水平上,Shannon信息指数(I)和Nei基因多样性(He)分别为0.971 9和0.556 7,表现出较高的遗传多样性;玉龙雪山不同海拔梯度上急尖长苞冷杉居群的遗传多样性有差异,且随着海拔梯度的递增,居群的遗传多样性除海拔3 350 m外,其他居群的遗传多样性随海拔的增加而增加,并在海拔3 700 m处达到最大值;急尖长苞冷杉居群间的基因流(N_m)为4.778 0,群体分化系数(Fst)为0.049 7,且基于AMOVA软件分析结果显示有3.55%的变异发生在居群间,表明急尖长苞冷杉群体间分化低,且大部分变异存在于居群内。研究结果表明海拔梯度对急尖长苞冷杉遗传多样性影响甚微。

Genetic Diversity of Abies georgei var. smithii at Different Altitudes in Jade Dragon Snow Mountain
Abstract:4 pairs of SSR markers were used to study the genetic diversity and genetic structure of 5 Abies georgei var.smithii populations at different altitudes in Jade Dragon Snow Mountain in the northwest of Yunnan Province.At the species level,Shannon information index (I) and Nei’s gene diversity (He) were 0.971 9 and 0.556 7,respectively,indicating that this species has high genetic diversity at the species level.Along the altitu?dinal gradients,the genetic diversity increased with the increase of elevation gradient, except for the altitude 3 550 m. The genetic diversity of the population at altitude 3 700 m was much higher than that of other popula?tions.And the gene flow Nm was 4.778 0.AMOVA analysis showed that only 3.55% of the total genetic variation occurred among populations,whereas 96.45% of the variance was within populations,which was in line with the coefficient of genetic differentiation (Fst = 0.049 7).The results indicated that the altitude gradient had lit?tle effect on the genetic diversity of Abies georgei var. smithii.
