吉林梅花鹿IGF-I基因启动子区单核苷酸多态性分析

摘    要:目的]对吉林梅花鹿IGF-I基因进行单核苷酸多态性分析,为梅花鹿产茸性状的分子育种提供新的分子生物学依据。方法]采用PCR-SSCP技术分析IGF-I基因在吉林左家梅花鹿群体中的多态性,并对IGF-I基因多态片段进行克隆、测序,确定多态位点的突变位置。结果]P1引物扩增的群体出现了AA、BB、AB和CC4种基因型,基因型频率分别为0.100、0.300、0.433和0.167,等位基因频率为0.317、0.517和0.166;P2引物扩增的群体出现3种基因型,分别用GG、TT和GT,基因型频率分别为0.500、0.333、0.167,等位基因频率为0.667和0.333。测序结果表明,在引物P1扩增片段中,在105~107处发生了AGT碱基的插入,在125和126处发生了T→A的突变,落在IGF-I基因的启动子区;引物P2扩增片段中,在236处有1个C→T的突变,落在IGF-I基因的启动子区。2个SNPs形成AG、AT、BG、BT、CG和CT6种单倍型,频率分别为0.239、0.077、0.311、0.206、0.117和0.050。结论]IGF-I基因在吉林左家地区梅花鹿群体中具有遗传多态性,发现了多个突变位点,然而这些碱基的突变是否影响到IGF-I基因的生物活性,还有待进一步研究。

Analysis of Single Nucleotide Polymorphisms at Promoter Region of IGF-I Gene in Cervus nippon from Jilin Province

Abstract:Objective] To analyze single nucleotide polymorphisms (SNPs) of IGF-I gene in Cervus nippon,provide a new molecular biology basis for the molecular breeding of antler producing traits. Method] PCR-SSCP analysis of IGF-I gene of sika deer population in Zuojia Area of Jilin Province was performed. After IGF-I gene polymorphism fragments were cloned and sequenced,the mutation locations of polymorphic loci were identified. Result] In primer P1 groups,four genotypes including AA,BB,AB and CC were found,the genotype frequencies were 0.100,0.300,0.433 and 0.167,respectively,and the allele frequencies were 0.317,0.517 and 0.166. In primer P2 groups,three kinds of genotypes,namely GG,TT and GT were found,the genotype frequencies were 0.500,0.333 and 0.167,respectively,and the allele frequencies were 0.667 and 0.333. The sequencing results showed that in the amplified fragments of primer P1,there was an AGT insertion at 105-107 nt and T→A mutations at 125 and 126 nt; in the amplified fragments of primer P2,there was a mutation of C236T. The haplotype frequencies of AG,AT,BG,BT,CG and CT were 0.239,0.077,0.311,0.206,0.117 and 0.050,respectively. Conclusion] In the population of sika deer at Zuojia Area of Jilin Province,IGF-I gene show genetic polymorphism and is found numerous mutations. However,whether these mutations affect the biological activity of the IGF-I genes remains to be further studied.
