低温预处理对大麦花药内源游离氨基酸和多胺水平的影响

摘    要: 以大麦花药离体培养反应敏感型和迟钝型品系为供试材料,测定了低温(3℃)预处理过程中花药内游离氨基酸和多胺的含量。试验结果表明,低温预处理 5 d时,所有供试材料的游离氨基酸总量水平均有所下降;15 d后,皆大幅度上升,敏感型材料的升幅比迟钝型材料的大。敏感型材料的游离脯氨酸、丝氨酸、精氨酸、丙氨酸含量水平明显比迟钝型材料的高。25 d低温预处理后,敏感型材料的多胺总量比 0 d时的明显提高,腐胺和亚精胺的含量升幅特别显著;迟钝型材料的多胺总量比0d时的低,亚精胺和精胺的降幅较为明显。依据上述结果,推测低温预处理导致药壁蛋白质降解成大量游离氨基酸,其中游离脯氨酸、丝氨酸、精氨酸、丙氨酸含量以及腐胺、亚精胺含量大幅度升高有利于小孢子脱分化。

Abstract:Using barley lines with low and high responding to anther culture as materials , changes of endogenous amino acid and polyamine contents in anthers were investigated. Cool-pretreatment of panicles for 5 days led to an decrease of total free amino acid content and 15 days of the pretreatment led to a big in- crease of the total content. In comparison with low responding lines, the total content of high responding line increased more obviously, especially for proline, serine, arginine and alanine. After 25 days of cool- pretreatment, the total content of polyamine was significantly higher than that of 0 day's treatment in high responding line and decreased in low responding lines. The contents of putrescine and spermidine increased more obviously in high responding line and decreased significantly in low responding lines. It can thus be suggested that large quantity of free amino acid decomposed from anther-protein and increase of free pro- line, serine, arginine, alanine and putrescine, spermidine benefits the microspores in starting its first mi- totic division.
