摘    要:[目的] 探讨ghrelin对蒙古绵羊脐静脉内皮细胞的血管内皮生长因子VEGF及其受体Flt-1 mRNA表达的影响。[方法] 试验分为4组:Ⅰ组,空白对照组;Ⅱ组,脂质体组;Ⅲ组(SCON),20 μmol/L正义寡核苷酸组;Ⅳ组(ASCON),20 μmol/L反义寡核苷酸组。上述各组均在培养24、36、48 h后,采用实时荧光定量方法检测VEGF及其受体Flt-1 mRNA表达情况的变化。[结果]VEGF mRNA在Ⅰ、Ⅱ组表达量无差异,具有较高的表达量,随着时间的延长无明显变化;在Ⅲ组VEGF mRNA 表达略有降低,但与Ⅰ和Ⅱ组间无显著性差异(P>0.05);Ⅳ组(反义寡核苷酸组)的VEGF mRNA表达量显著下降(P<0.05),并且随着时间延长逐渐下降。VEGF受体FLT-1 mRNA的表达与VEGF相似。[结论]反义抑制ghrelin对VEGF及其受体Flt-1的mRNA表达有下调作用。

Effects of Ghrelin Antisense Inhibition on VEGF and Its Receptor Fit-1 mRNA Expression

Abstract: Objective ] This study was to investigate the effect of VEGF and its receptor Fh-1 mRNA expression in Mongolia sheep' s umbilical vein endothelial cells by ghrelin antisense inhibition. Method] Experiments were divided into 4 groups:group Ⅰ (blank control group) ; group Ⅱ (liposome group) ; group Ⅲ(SCON group: 20 μmol/L sense oligonucleotide) ; group Ⅳ (ASCON: 20 μmol/L antisense oligonucleotide). VEGF and its receptor Fh-1 mRNA expression changes were detected by using real-time fluorescence quantitative detection after 24, 36 and 48 h. Result] The expression of VEGF mRNA in group Ⅰ, group Ⅱwere insignificantly different at higher expression levels, and did not change significantly with the time; the expression of VEGF mRNA in group Ⅲ assumed a slight decrease, but there were no significant differences between group Ⅰ and group Ⅱ (P 〉 0.05 ), the expression of VEGF mRNA in group Ⅳ ( antisense oligonucleotide group ) decreased significantly ( P 〈 0.05 ) ; the expression of VEGF receptor FLT-1 mRNA was similar to that of VEGF Conclusion ] Antisense inhibition ghrelin has a downward effect to the expression of VEGF and its receptor Fh-1.
