欧亚大陆水青冈种群遗传多样性对比分析

摘    要:该文应用凝胶电泳法测定中国水青冈3个种,即米心水青冈、巴山水青冈和亮叶水青冈的种群遗传多样性;分析了各物种种间、种内的遗传变异和亲缘关系. 同时与欧洲水青冈进行了对比,探讨不同环境下水青冈的地理变异及其变异的机制. 所测定的酶系统包括:过氧化物酶(PX1和PX2)、磷酸葡萄糖脱氢酶(PGD)、酸性磷酸化酶(ACP和ACP2)、超氧化物歧化酶(SOD)、谷氨酸草酰乙酸转氨酶(GOT1、GOT2和GOT3)、异柠檬酸脱氢酶(IDH)、磷酸果糖异构酶(PGI)、甲基萘醌还原酶(MNR)、葡萄糖磷酸变位酶(PGM1和PGM2)和苹果酸脱氢酶(MDH1和MDH2)10种酶系统. 研究结果表明,各物种特有等位基因是分析遗传多样性的关键,不同季节气候和不同地理条件下,水青冈种内基因频率和等位基因表达也不同. 亮叶水青冈和米心水青冈都含有较多的特有等位基因,是相对独立和分布范围广泛的树种;亮叶水青冈与巴山水青冈的亲缘关系最近,而米心水青冈与亮叶水青冈和巴山水青冈的亲缘关系都远.但这几种与欧洲水青冈相比,又相对具有更近的亲缘关系.

Abstract:Based on the analysis of geo-historical data and gel electrophoresis measurement, the genetic diversity and genetic structure of 11 populations of three species were studied, which included Fagus engleriana, F. pashanica and F. lucida. The genetic diversity and genetic structure of 7 Eurasian beeches with the changes of latitude, longitude and altitude were discussed. Ten allozymes including peroxydase (PX- 1and PX- 2), phosphogluconate dehydrogenase (PGD), acid phosphatase (ACP- 1and ACP- 2), superoxide dismutase (SOD), glutamate oxaloacetate transaminase (GOT- 1, GOT- 2 and GOT- 3), isocitrate dehydrogenase (IDH), phosphate glucose isomerase (PGI), menadione reductase (MNR), phosphoglucomutase (PGM- 1 and PGM- 2) and malate dehydrogenase (MDH- 1 and MDH- 2) were selected to analyze the genetic similarity, genetic diversity and genetic distance of the beeches distributed both in Europe and Asia. The results show that the allelic frequency varies with seasonal and geographic conditions, and both F. lucida and F. engleriana contain many special allelic genes. It can be concluded that the two species are widely distributed but relatively independent on the other species; F. lucida and F. pashanica have a close genetic relationship, while F. engleriana has a much far relationship with F. lucida and F. pashanica. F. sylvatica has a far relationship with all the three Chinese species. Finally, the allozyme spectrum and dendrogram of Eurasian beeches were made.
