家蚕核型多角体病毒酪氨酸蛋白磷酸酯酶基因核苷酸序列分析

摘    要:从家蚕核型多角体病毒中国镇江株 (BmNPV ZJ)基因组DNA中克隆出酪氨酸蛋白磷酸酯酶基因 (ptp) ,该基因的编码部分由 5 0 7个核苷酸组成 ,其中A为 16 3、C为 99、G为 113、T为 132 ,G +C含量约为 4 2 %。根据其核苷酸序列推演的蛋白质由 16 8个氨基酸残基组成 ,其中含有酪氨酸蛋白磷酸酯酶催化活性区的 11个氨基酸“HC”基序。该基因与苜蓿银纹夜蛾核型多角体病毒 (AcMNPV)ptp和BmNPV T3株 (日本 )的ptp核苷酸序列的同源性分别为 96 8%和 98 2 %。BmNPV ZJ酪氨酸蛋白磷酸酯酶 (BmNPV ZJPTPase)的氨基酸全序列与AcMNPV、BmNPV T3、芹菜夜蛾核型多角体病毒 (AfMNPV)PTPase和黄杉毒蛾核型多角体病毒 (OpMNPV)PTPase 1的氨基酸全序列的同源性分别为 97%、97 6 %、96 %和 6 0 % ,而与OpMNPVPTPase 2的同源性仅为 2 0 %。在NCBI数据库中查找BmNPV ZJPTPase的同源性序列 ,查找到的 5 99381个序列中发现至少有 14种mRNA加帽酶其N端部分存在PTPase催化活性区的“HC”基序 ,但其氨基酸全序列的同源性只有 31%~ 32 %。该基因序列已被GenBank数据库收录 ,登录号为AF316 871

Abstract:A protein tyrosine phosphatase (PTP)gene was cloned from the genome of Bombyx mori nuclear polyhedrosis virus,Zhenjiang strain (BmNPV ZJ) and sequenced.The coding region of the gene composed of 507 nucleotides with the base count of 163 A,99 C,113 G,132 T and a G+C content of 42%.The 168 amino acid sequence deduced from the nucleotide sequence contained the conserved 11 amino acid "HC" motif(residues 117~127) charactering PTPase family.The BmNPV ZJ PTPase gene shared 96.8% identity in nucleotide sequecce with its homologue in Autographa colifornica multinucleocapsid nuclear polyhedrosis virus (AcMNPV) and 98.2% identity with BmNPV T3(Japan) homologue.Its amino acid sequence shared identities of 97%,97.6%,96% and 60% respectively with its homologues in AcMNPV,BmNPV T3,AfMNPV( Anagrapha falcifera multinucleocapsid nuclear polyhedrosis virus)and with OpMNPV ( Orgyia pseudotsugata multinucleocapsid nuclear polyhedrosis virus) PTPase 1.However,the identity of BmNPV ZJ PTPase to OpMNPV PTPase 2 was only 20%.By searching homologous sequence to BmNPV ZJ PTPase from the 599,381 amino acid sequences in NCBI data base,at least 14 mRNA capping enzymes were found to contain the PTPase active site "HC" motif at the region of their N terminals,but their identity in complete amino acid sequence was only 31%~32%.The newly determined nucleotide sequence has been submitted to GenBank and the accession number is AF316871.
