摘    要:以南方四季杨叶片作为受体材料,分别设置梯度实验,研究了Kan(卡那霉素)选择压以及遗传转化过程中影响转化效率的5个主要因素(预培养时间、浸染时间、共培养温度、共培养时间和乙酰丁香酮浓度),得出30mgL-1的Kan浓度作为南方四季杨遗传转化过程中的选择压。高效遗传转化体系为:叶片预培养3 d;OD600=0.4的菌液浸染10 min~15 min;浸染前菌液中加入AS(乙酰丁香酮)300 umol·L-1;22℃~25℃条件下暗处共培养3d。

Abstract:Five main factors affecting transformation of Populus deltoides × P.nigra cv.Chile were studied by Agrobacterium tumefaciens-mediated infecting and experimented with various gradients,such as pre-culture time,infection time,co-culture temperature,co-culture time,acetosyringone(AS) concentration in bacterium fluid;In addition,Kan selection pressure was researched.The result showed that 30 mg·L-1 Kan concentration were suitable.An effective protocol was as follows: 3 days for pre-culture of leaf blades,10 minutes~15 minutes for infection in the OD600=0.4 Agrobacterium tumefaciens bacterium fluid;adding As 300 μmol·L-1 in the bacterium fluid dosage before infection,and 3 days for co-culture in dark under the condition of 22℃~25℃.
