籼稻离体胚培养中体细胞胚胎发生的研究

摘    要:成熟稻胚在含2,4-D的MS培养基上产生胚性愈伤组织以及在胚性愈伤组织表面形成许多瘤状突起的原胚状体,组织学研究表明在诱导培养的不同时期,胚性愈伤组织的细胞分裂产生单核胚性细胞、2细胞、4细胞、多细胞及球形胚。随后移到不含2,4-D的分化培养基上培养,胚性愈伤组织产生与水稻合子胚结构相似的成熟体细胞胚状体。结果表明:2,4-D是胚性愈伤组织诱导的必要激素,但抑制球形胚的发育成熟。不同激素处理结果:Br明显抑制植株再生。研究生理生化指标结果表明:在愈伤组织生长分化过程中不同激素处理Br抑制氧化物酶活性;体细胞胚状体在形成、成熟阶段,酶活性及酶带相应增强和增多;当胚状体萌发形成植株时,酶活性及酶带相应下降和减少,随诱导培养时间延长植株再生率降低。

Studies on Somatic Embryogensis of Indica Rice from Dehulled Seed Culture in Vitro
Abstract:The formation of embryonic cellus and somatic cmbryogensis were establisned from dohulled mature seeds. The results of histologicai studies showed that embryoids could be derived from single cells of embryonic callus. Having the same structure as a zygotic embryo of rice, a full-developed embryoid possessed the major embryonic parts,such as scutellum, celeoptile and coleorhiza. Meanwhile, abnormal embryoids were aslo observed in the experiment.Based on the embryogensis, the celeoptile was considered as the histologicai index. The increase of peroxidase activtiy was followed by the development of somatic embryoids. The peroxidase and isoperoxidase were considered as the physiological and biochemical index. The activity of peroxidase was inhibited when Brassinolide was added to the inducing medium, and the number of plantlets decreased. It was necessary for the development of somatic embryoids that the medium contained 2,4-D, with an optimum concentration of 1 ppm. However, differentiation would be inhibited when the globe embryoid was kept culturing on the medium containing 2,4-D, due to that the Br and 2,4-D inhibited peroxidase activity . Generally, differentiation of embryonic callus was affected by the duration of this process. It was pointed out that fifteen days would be the best For differentiation.
