| 摘 要： | 给6只健康杂种细毛羊,采用磺胺嘧啶钠注射液,以70mg/kg,一次快速静脉推注,给药后不同时间采血,用重氮化——偶合比色法测定SD浓度。经模型选择,药时变化符合无吸收因素二室模型(=Ae~(-αt)+Be~(-βt)),按此模型计算动力学参数:半衰期(T1/2β)为2.274±0.967小时;有效血浓度维持时间(TCP)为3.116±0.767小时。表明SD在该品种绵羊体内代谢较快,应属短效磺胺。建议临床应用以每公斤体重100毫克静注,间隔12小时。因为据此算得稳态平均血浓度(C_∞)为5.869mg/100ml,在有效血浓度以上(最低有效血浓度为5mg/ml);蓄积因子R为1.02,符合R在1.4左右。如果以每公斤体重70mg静注,应每4小时给药一次,为克服用药过频,应与符合该品种羊的中效、长效或口服磺胺药配合应用。

| Abstract: | Sulfadiazine(SD) was administered intravenously at a dose of 70mg/kg to six crossbred sheep. Blood samples were taken from the jugular vein at 0.08,0.25,0.5,1,2,3,4,6,8houts postadministration and blood concentration were measured by Diazotization Coupling Colorimetry. The serum concentration versus time date were analysed, using statistical moment theory. The i/v data were also fitted by open two-compartment model.Pharmacokinetic parameters were calculated (biological half-life(t~1/_2β)2.274±0.967hours; therapeutic concentration period(TCP)3.116±0.767hours). It was shown that SD should belong to the range of short effecting Sulfonamides in metabolism of the sheep. It is suggested that sheep should be given SD in jugular vein at 100mg/kg body weight every 12 hours or at 70mg/kg body weight every 4 hours, and SD should be coordinated with middle or long effecting Sulfonamides in order to avoid the frequent use of the drug. |
