桑树茎尖嫁接技术研究

摘    要:在无菌条件下将桑籽接种至含MS无机盐与0.2%凝胶剂(Gelrite)的培养基上,于26℃每日16h、10001x照明下培养;2周后以激素处理茎切段,转培于蛭石培养基,10~12d苗茎围度达8mm时,以长2cm的切段为砧木,以707等桑品种的1~2mm茎尖作接穗,嫁接成活率达75%~80%。增大砧木苗基的围度是取得桑树茎尖嫁接成功的关键。

Abstract:A procedure of shoot-apex grafting in vitro of mulberry is described. Mulberry seeds were inoculated on Murashige and Skoog(MS) medium solidified with 0.2%Gel-rite under aseptic conditions and incubated in a chamber at 26℃,16h daily exporsure to1000lx illumination for two weeks. Sboots were excised with their bases soaked in hor-mone solution and transferred to vermiculite medium, incubated in the chn1ber at 26℃and constant darkness for l0~12 days, When the circumference of the seedling stemsreached about 8mm,they were served as rootstocks for shoot-apex grafting in vitro. Anaverage frequency of 75%~80%of successful shoot-apex grafting of several major com-mercial varieties was achieved by using 2 cm segments from the seedlings as rootstocksand l~2mm shoot apices from adult plants as scions,It is concluded that thickening thecircumference of rootstock stems is the most critical factor for successful shoot-apexgrafting in vilro of mulberry.
