多汁乳菇多糖对酒精性损伤小鼠心脏及肾脏的抗氧化作用

摘    要:以超声波辅助热水浸提并去蛋白得到的多汁乳菇精多糖(refined polysaccharide from Lactaruis volemus Fr.,RPLV)为试验材料,探究RPLV对急性酒精性损伤小鼠心脏、肾脏的保护作用。将55只小鼠随机分成空白组、阳性对照组、模型组、RPLV高和低剂量组(400 和100 mg·kg-1(bw)),连续给药30 d,每组小鼠断食12 h后,除空白组,其余组按照12 mL·kg-1的剂量灌胃50%乙醇,建立急性酒精性损伤模型。12 h后,取小鼠心脏、肾脏,检测其丙二醛(malondialdehyde, MAD)、还原型谷胱甘肽(glutathione, GSH)含量以及过氧化氢酶(catalase, CAT)、超氧化物歧化酶(superoxide dismutase, SOD)和谷胱甘肽过氧化物酶(glutathione peroxidase, GSH-Px)的活性。结果表明,心脏、肾脏的RPLV各剂量组与模型组相比,MDA含量均显著或极显著降低(P < 0.05或P < 0.01),而SOD、GSH-Px、CAT活性和GSH含量均显著或极显著升高(P < 0.05或P < 0.01)。可见RPLV可以通过抗氧化作用在一定程度上保护由酒精引起的心脏、肾脏损伤。

Abstract:To explore the protective effects of refined polysaccharide from Lactaruis volemus Fr. on acute alcoholic injury to the heart and kidney of mice, we used refined polysaccharide from Lactaruis volemus Fr., which was gained after hot water extraction, assisted by the ultrasonic as well as deproteinization, as the material. Fifty-five mice were randomly divided into groups as follows: normal control group, model group, positive control group, RPLV high and low dose group (400 and 100 mg·kg-1(bw)). Continuous administration for 30 days, the mice in each group were deprived of food for 12 h. Except the normal control group, the other groups were given 50% ethanol by gavage at a dose of 12 mL·kg-1 to establish an acute alcoholic injury model. After alcohol treatment for 12 h, the mice heart and kidney were removed to determine malondialdehyde (MAD), glutathione (GSH) content, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities. As results, compared with the model group, the RPLV dose groups in heart and kidney showed significant or extremely significant decrease in MDA content (P < 0.05 or P < 0.01), while the SOD, GSH-Px, CAT activities and GSH content were significantly or extremely significantly increased (P < 0.05 or P < 0.01). In conclusion, RPLV can protect heart and kidney injury caused by alcohol to some extent through antioxidant effect.
