摘    要:高质量的DNA是进行林木分子生物学研究的基础与关键。该研究利用改良的CTAB法提取垂柳基因组DNA,并与常规CTAB法、CTAB-硅珠裂解法、SDS法和试剂盒法进行了比较。琼脂糖凝胶电泳检测显示,改良CTAB法提取的DNA浓度和纯度较高,质量优于其他4种提取方法。同时利用改良CTAB法提取了柳树16个自然种及杂交种基因组DNA,并经SSR-PCR扩增反应验证,表明改良CTAB法可作为一种柳树基因组DNA提取的通用方法。

A modified method of DNA extraction from Salix

Abstract:High quality DNA is critical in the study of forest molecular biology. In the present study, a method of DNA ex- traction from Salix babylonica was modified and compared with regular CTAB method, CTAB-silicon bead cracking method, SDS method and DNA extraction kit method. Agarose gel electrophoresis result showed that the concentration and purity of extracted DNA were superior to the other four methods by using modified CTAB method. Through the validation of extrac- tion from 16 species and hybrids of Salix and amplification of SSR-PCR, we concluded that the modified CTAB method could be universal for genomic DNA extraction of Salix sp.
