| 摘 要： | 目的]观察九香虫含药血清对人结肠癌细胞SW480增殖及凋亡相关因子FADD、p53表达的影响,初步探讨其诱导结肠癌细胞凋亡的作用机制。方法]20只雄性SD大鼠按随机数字表法分为2组（生理盐水对照组和九香虫含药血清组）,每组10只。其中,九香虫含药血清组给予九香虫煎剂灌胃5 d后,制备九香虫含药血清,按不同浓度体外作用于SW480细胞72 h。CCK8细胞增殖试验检测其对SW480细胞增殖的抑制作用,流式细胞术检测其对SW480细胞凋亡的诱导作用,Western blot法、荧光定量PCR法检测细胞中FADD、p53蛋白及mRNA的表达。结果]各浓度九香虫含药血清对SW480细胞生长均有抑制作用,且明显促进其凋亡,凋亡率明显高于对照组（P〈0.01）。Western bolt及荧光定量PCR结果表明,九香虫含药血清干预可使FADD表达明显增强,p53表达明显减弱。结论]九香虫含药血清可诱导人结肠癌细胞凋亡,并影响凋亡相关因子p53、FADD的表达,从而达到抗肿瘤作用。
