甘蓝型油菜EMS诱变后代农艺性状观察及分子检测

摘    要:利用不同浓度甲基磺酸乙酯(EMS)溶液处理甘蓝型油菜常规品种川油20和渝黄1号选系种子,结果表明:EMS对成苗率的抑制作用随着浓度的增加而增强,浓度1.0%为半致死量。M1中出现叶片卷曲、皱缩、黄化、双茎、扁茎和植株矮化等变异性状;M2中筛选到双茎、长角、多分枝等变异性状。对M2长角果等产量性状变异株进行考察,结果表明变异株角果长度(果身和果喙)高于对照,有效分枝数、有效角果、角果粒数和千粒重高于对照,一次分枝高度低于对照。对变异株进行SSR分析表明,变异株与对照在电泳条带上表现出多态性,进一步证明了变异的真实性。

Abstract:In order to establish the variation population,the seeds of pure lines "Chuanyou 20" and "Yuhuang No.1"(Brassca napus L.) were treated with EMS at different concentrations.The results showed that the survival rate was decreased with the increase of EMS concentration,and those of 1.0% was the half-lethal dose for two materials.Variation with abnormal appearance of curled leaf,yellow leaf,the double stems and dwarf plant were observed in the M1 population.Variant plants with long-pod,branches,aggregate silique and double stems were screened in M2.In vestigation of the agronomic traits of variant plants,such as yield traits of long-pod in M2,indicated that the pod(pod body length and beak length) of variant plants was longer than control;the numbers of effective branches and pods per plant,seeds per pod and 1000-seed weight were also higher than control,whereas the height of primary branches was lower than the control.The existence of the variation was further proved by molecular marker due to the detection of polymorphic SSR between variant plants and control.
