摘    要:已知许多植物抗病基因的蛋白质产物具有保守的结构域 ,如NBS、LRR、TM等。根据烟草的N基因、亚麻的L6基因和拟南芥的RPS 2基因的保守序列设计了 1对简并引物 ,并以应县小黑豆基因组DNA为模板 ,PCR扩增获得 16个不同的RGA片段 ,大小在 470~5 45bp ,且不同程度地含有抗病基因的保守序列 ,如GGVGKTT、GSRII、GLPL等。与已知大豆、棉花RGA进行比较 ,发现该试验获得的大豆RGA和用相同引物扩增获得的棉花RGA核酸序列以及相应的氨基酸序列之间同源性较低 ,与已经克隆的大豆RGA及其氨基酸序列之间相似性较高。在分子水平揭示了大豆及棉花RGA的起源及其进化。

Abstract:There are conserved domain in protein products of the many known gene, such as Nucleotide Binding Site (Nucleotide Binding Site, NBS), LRR(leucine rich repeat, LRR) and TM(transfer membrane,TM).In this study, the degeneration primer was designed based on the conserved NBS of the genes of N,L6,and RPS2.Specific fragments of 517bp were amplified from genomic DNA of Soybean. The 16 fragments were obtained and they were sequenced. Moreover, They were compared with cotton RGAs and cloned soybean RGAs by DNA man software. The results showed the cloned RGAs in this experiment had high similarity to the known soybean RGAs. But it low similarity to cotton RGA were obtained by same primers. The results revealed molecular mechanism for evolution of soybean RGA and cotton RGA.
