西瓜种子大小形状相关QTL分析

摘    要:以黏籽西瓜(Citrullus lanatus ssp. mucosospermus)‘PI186490’和普通栽培种西瓜(Citrullus lanatus ssp. vulgaris)‘LSW-177’为亲本,利用F2群体对种子大小和形状的QTL进行分析,构建了1张包含195个CAPS标记,并与染色体一一对应的11个连锁群的西瓜种子相关性状遗传连锁图谱。该图谱覆盖基因组长度2 029.8 cM,标记间平均距离10.46 cM。共检测到与种子相关性状QTL 23个,分布在连锁群LG1、LG2、LG3、LG5、LG6、LG8和LG11上,LOD值介于2.5208 ~ 36.0847之间,可解释2.2014% ~ 28.8950%的表型变异率。贡献率 > 17%的QTL位点4个,其中种子长度、种子宽度、种子厚度及种子百粒质量各1个。连锁群LG6定位到10个QTL,包含3个种子长度QTL、4个种子宽度QTL、1个种子厚度QTL及2个种子百粒质量QTL,4个贡献率 > 17%的QTL位点全部定位在连锁群LG6上,其中种子宽度和种子百粒质量主效QTL共同定位在标记WⅢ6-503和WⅢ6-502之间。

Abstract:The inheritance of seed characteristic was studied by using the populations derived from the Egusi watermelon PI186490 and the cultivated watermelon LSW-177. A F2 genetic linkage map which included 195 CAPS markers was constructed. The map contained 11 linkage groups which corresponded with the chromosome and spanned 2 029.8 cM with a mean marker interval of 10.46 cM. Twenty-three QTLs for the seed traits were detected on LG1,LG2,LG3,LG5,LG6,LG8,LG11 and explained 2.2014%–28.8950% of the phenotypic variation. Of all the QTLs,4 QTLs for seed length,seed width,seed thickness and 100-seed weight explained over 17% of the phenotypic variation. Ten QTLs were identified on LG6,which contain 3 QTLs for seed length,4 QTLs for seed width,1 QTL for seed thickness and 2 QTLs for 100-seed weight. Four QTLs which explained over 17% of the phenotypic variation were also located on LG6 and QTL for seed width and 100-seed weight were both located on the two CAPS markers WⅢ6-503 and WⅢ6-502.
