家蚕三磷酸腺苷结合盒转运子B亚族基因Abcb6的克隆与序列分析及组织表达

摘    要:三磷酸腺苷结合盒转运子B亚族成员ABCB6(ATP-binding cassette transporter subfamily B member 6,ABCB6)是一个重要的半分子转运子,在哺乳动物中主要参与细胞内卟啉类化合物的转运和铁离子平衡的调节。利用人的ABCB6蛋白的氨基酸序列在家蚕EST数据库中进行TBLASTN检索,将检索到的EST序列进行电子延伸和重叠群拼接,根据拼接结果设计引物进行RT-PCR扩增,成功克隆了家蚕Abcb亚族基因第1个成员BmAbcb6的完整开放阅读框。用生物信息学方法检测发现,BmAbcb6有16个外显子和15个内含子,编码850个氨基酸残基,分子质量96.35 kD,等电点8.23;BmAbcb6含有1个核苷酸结合结构域和10个α螺旋构成的跨膜结构域,属家蚕Abcb亚族成员。半定量RT-PCR方法分析BmAbcb6在家蚕5龄第3天幼虫不同组织中均有较高表达,其中以精巢中的表达水平最高。研究结果为进一步分析该基因的功能奠定了基础。

Abstract:ATP-binding cassette subfamily B member 6,ABCB6,is a important half-molecule transporter.In mammals,ABCB6 is shown to be functionally active in cellular efllux of certain porphyrins from cells and to be involved in regulation of iron homeostasis.The amino acid sequence of human ABCB 6 protein was used to conduct TBLASTN search against silkworm(Bombyx mori) EST database.The obtained ESTs were extended in silico and assembled into a consensus sequence,based on which primers were designed for RT-PCR amplification.As a result,the intact open reading frame of BmAbcb 6,the first silkworm Abcb subfamily gene,was cloned successfully.Bioinformatics analysis showed that BmAbcb 6 has 16 exons and 15 introns,encoding 850 amino acid residues with a predicted molecular mass of 96.35 kD and pI 8.23.BmAbcb6 protein contains one nucleotide binding domain and one transmembrane domain containing 10 α-helices.Semi-quantitative RT-PCR analysis showed that BmAbcb 6 gene was highly expressed in various tissues of the 5th-instar day-3 larvae of silkworm,with highest expression in testis.This result paves the way for better understanding the function of BmAbcb 6 gene.
