大白菜小孢子植株的倍性变异及倍性鉴定方法的研究

摘    要:对大白菜小孢子植株群体的倍性变异进行观察,并以染色体计数法的鉴定结果为依据,研究利用DNA流式细胞仪测定法、气孔保卫细胞特征、形态学特征鉴定大白菜小孢子植株倍性的可靠性。结果表明,小孢子植株是倍性水平不同的混合群体,但不同基因型各倍性植株所占比例不同;DNA流式细胞仪鉴定法的准确率达94.44%,可用于植株群体的倍性鉴定;植株形态鉴定法可用于成株倍性鉴定;气孔特征鉴定法的应用有待进一步探讨。

Abstract:In this research the ploidy variation of microspore-derived plants in Chinese cabbage was investigated. The results showed that regenerated populations from microspore are usually mixed ploidy and the distribution of haploid, diploid, trploid, tetraploid and chimaera is various in each genotype material. Based on the result of chromosome counting, the authenticity of ploidy determination of microspore-derived plants in Chinese cabbage were further evaluated using several different approaches. The accuracy of ploidy determination measured with DNA flow cytometry reached 94.44 % ,which indicated DNA flow cytometry could be used to identify ploidy level. The method of morphological characteristics could be used to estimate the ploidy level of ripe planflets. The characteristic value of stomata is difficult to regard as the index of ploidy qualificaiton,it needed to be further investigated.
