摘    要:为了提高紫色硫细菌283-1的生物量,对其培养基组分进行优化。采用单因子试验、Plackett-Burman设计法、最陡爬坡试验及中心组合设计响应面分析,研究了乙酸钠、碳酸氢钠、氯化铵、硫化钠、硫酸镁和无机盐6个因子对菌株283-1生物量的影响。结果表明,乙酸钠和硫化钠是影响该菌株生物量的主要因素,并获得了最佳培养基配方:乙酸钠1.45g·L-1,碳酸氢钠0.75g·L-1,氯化铵4.5g·L-1,硫酸镁0.5g·L-1,硫化钠1.05g·L-1,氯化钙0.19g·L-1,氯化钾0.34g·L-1,氯化钠50g·L-1,VB1220μg·L-1,无机盐溶液1mL·L-1。优化后菌株283-1的生物量提高了6倍,结果与理论预测值相近。

Abstract:In order to improve the biomass,medium composition of the strain 283-1 of purple sulfur Bacterium was optimized.Effects of the six factors CH3COONa,NaHCO3,NH4Cl,Na2S,MgSO4 and inorganic salt on the biomass were evaluated by using single-factor test,Plackett-Burman design,the steepest ascent experiment and central composite design combined with response surface analysis in this work.The results showed that sodium acetate and sodium sulfide were the most significant factors and the optimal media were as following: CH3COONa 1.45 g·L-1,NaHCO3 0.75 g·L-1,NH4Cl 4.5 g·L-1,MgSO4 0.5 g·L-1,Na2S 1.05 g·L-1,CaCl2 0.19 g·L-1,KCl 0.34 g·L-1,NaCl 50 g·L-1,VB12 20 μg·L-1 and inorganic salt 1 mL·L-1.With the optimized media,the biomass improved 6 times compared with the control and the value was in agreetment with the predicted.
