| 摘 要： | 以猪脏器为材料,应用TRIpure Reagent总RNA提取试剂盒提取脏器总RNA,发现直接按照试剂盒说明书操作提取的RNA效果不理想。通过反复试验,改进了提取过程中的部分操作步骤。结果表明:改进后与改进前相比,提取RNA的纯度和得率在0.01水平上有差异;改进后提取的RNA经RT-PCR扩增猪"βactin基因,电泳结果显示,扩增条带清晰、特异,说明提取的RNA完全满足后续分子生物学试验的要求。

| Abstract: | In this study,commercial TRIpure Reagent Total RNA Extraction Kit was used in RNA extraction from porcine viscera samples.We found the result was not satisfied if the operation was followed according to the manufacturer's instruction completely.After repeated tests,some modifications were tentatively adopted.And the comparative experiment result showed the extracted RNA purity and yield were significantly improved(P<0.01) following the modified instruction.With these RNAs,β-actin was amplified and the electrophoresis result showed clear,specific and integral bands.This finding showed that the extracted RNA by modified method can meet the requirement of the molecular research. |
