二花脸猪与大白猪肝脏生长激素受体基因表达的发育性变化

摘    要: 选刚出生的雄性纯种二花脸猪和雄性纯种大白猪各 2 8头 ,前者按中国地方猪种饲养标准饲养 ,后者按美国NRC标准饲养。测定不同发育阶段平均日增重 (BWG) ,并分别于 0、3、2 0、30、90、12 0、180日龄宰杀取样 ,用相对定量RT PCR方法 ,以 18SrRNA作内标 ,测肝脏生长激素受体 (GHR)mRNA丰度。结果发现 ,(1)从出生到 180日龄大白猪体重和BWG均显著高于二花脸猪。 (2 )大白猪和二花脸猪肝脏GHR基因表达发育性变化趋势相同 ,肝脏中GHR基因表达在出生时较低 ,随后表达逐渐增加 ,12 0日龄时达峰值 ,随后维持在较高水平 ,其发育性变化与BWG显著相关 (二花脸猪R =0 .6 8,P <0 .0 1,大白猪R =0 .96 ,P <0 .0 1)。双因子分析表明 ,大白猪肝脏GHR的表达极显著高于二花脸猪 (P <0 .0 1) ,其中从初生到 30日龄品种间差异不明显 ,在 90日龄到 180日龄大白猪肝脏GHR表达明显高于二花脸猪。提示 ,肝脏中GHR基因表达具有显著的年龄依赖性和明显的品种特征 ,肝脏中GHR基因表达对动物的生长有重要影响。

Abstract:Pure bred male Large White (LW) and Erhualian (EHL) piglets from birth (0 day) to 180-day-old were used in the present study to investigate the developmental changes of GHR mRNA expression in the liver. The animals were fed according to the breeding standards of USA NRC and Chinese Local Pigs respectively. Four piglets of each breed were slaughtered at 0, 3, 20, 30, 90, 120, 180 days respectively for tissue sampling. The liver total RNA was extracted to determine the abundance of liver growth hormone receptor messenger ribonucleic acid (GHR mRNA) by relative quantitative RT-PCR with 18S rRNA as internal standard. The body weight and average daily body weight gain (BWG) of LW at each age were higher than those of EHL. The developmental tendency of liver GHR mRNA expression in EHL and LW pigs were similar, from birth to 120 days the GHR mRNA abundance increased with age, but from 120 days onward, the expression of GHR mRNA kept pleatuea. The expression of GHR in liver was correlated with BWG (R=0.96 in LW; R= 0.68 in EHL). The results suggest that the liver GHR gene expression is affected by age and genotype, and plays an important role in animal development.
