摘    要:目的]研究了新疆白鲜愈伤组织诱导及其生长动态,为从中提取药物活性成分奠定基础,同时是解决资源贫乏的一条新途径。方法]以幼叶、真叶、下胚轴为外植体,研究不同培养基、激素配比、碳源对新疆白鲜出愈率的影响以及愈伤组织生长动态变化。结果]幼叶在激素浓度为6-BA 2.0 mg/L+2,4-D 0.5 mg/L的MS和B5培养基出愈率分别为83.0%和93.3%,且在低盐B5培养基上诱导率较高;不同外植体幼导率为子叶>下胚轴>真叶,子叶的诱导较快,诱导率较高;蔗糖作为碳源时效果较好,诱导率为100%;愈伤组织生长的倍增曲线揭示继代培养时,第5~17天为快速增长期,且在暗处生长较好,继代周期20 d较为合适;不同激素浓度对愈伤组织的质量亦有较大的影响。结论]y1y、2激素组合诱导出的愈伤组织虽较致密,但在体胚发生中均未达到理想效果,也未能进一步分化。将其转入含有激动素KT 3.0 mg/L+NAA 0.1 mg/L的MS培养基中,发现愈伤组织在光下变黄绿色,生长也较紧凑,可诱导出胚状体。

Callus Induction and Growth Dynamic of Dictamnus angustifoliu
Abstract:Objective] Callus induction and the growth dynamic of Dictamnus angustifoliu had been studied,which provided a basis to extract active constituents,and a new way to deal with natural resources shortage.Methods] Taking euphylla,cotyledon and hypocotyledonary axis as explants,the influences on the callus induction rate and the growth dynamic of Dictamnus angustifoliu in different medium,hormone matching and carbon sources were investigated.Results] The induction rate were 83.0% and 93.3% in MS and B5 mediums with 6-BA 2.0 mg/L +2,4-D 0.5 mg/L.And the induction rate was higher in B5 medium with low salt.The induction rates of different explants were as follows: cotyledon>hypocotyledonary axis>euphylls.Cotyledon inducted faster and got higher induction rate.Sugar was the best carbon source,with the induction rate of 100%.The study of the multiplication curve of the growth of callus revealed that from the fifth to the seventeenth day was the fast epacme in secondary culture.It grew better in darkness,and the subculture cycle was 20 days.Different hormone concentrations had great influences on the quality of callus.Conclusion] The grouping of hormones y1 and y2 inducted more compact calluses,but hadn't got desired results and couldn't differentiate further.Transferred to MS medium with KT 3.0 mg/L +NAA 0.1 mg/L,callus turned flavovirens in light,grew compactly and embryoids could be inducted.
