青杄PwVQ1基因的克隆与表达

摘    要:以青杄cDNA为模板克隆得到青杄VQ基因,命名为PwVQ1。序列分析表明,PwVQ1基因的开放阅读框为819 bp,编码272个氨基酸。生物信息学分析发现,PwVQ1编码的蛋白理论分子量为69.05 k Da,PI值为5.03,为非跨膜的亲水性蛋白。PwVQ1具有VQ家族典型的Fxxx VQx LTG结构域。实时荧光定量PCR分析结果表明,PwVQ1在青杄的根、茎、叶、花粉、果实、种子中都有表达,在根和叶中表达量十分显著;PwVQ1在不同逆境处理下均有响应。4℃冷胁迫下,PwVQ1的表达量在3 h时先下调,在6 h时明显上调,达到8倍左右,在12 h时表达量再次下调。42℃高温胁迫下,PwVQ1的表达量下调。在盐胁迫下,干旱胁迫和ABA处理下,PwVQ1的表达量均上调。推测PwVQ1参与干旱、ABA、高温、低温、盐胁迫等非生物胁迫的响应。

Abstract:VQ gene was cloned from Picea wilsonii cDNA library,named PwVQ1.By sequence analysis,the open reading frame of PwVQ1 gene was 819 bp,encoding 272 amino acids.By bioinformatics analysis,the protein encoded by PwVQ1 had a theoretical molecular weight of 69.05 kDa and theoretical isoelectric points(pI)value of 5.03, belonging to a non-trans-membrane hydrophilic protein.The results of tissue specificity experiments showed that PwVQ1 was expressed in different tissues,while highly expressed in leaf and stem.Besides, PwVQ1 participate in various abiotic stresses to different de-grees.The expression of PwVQ1 was down regulated at 3 h,up to 8 times at 6 h,and decreased again at 12 h under 4℃stress while the expression of PwVQ1 was down regulated under 42℃stress.Besides,PwVQ1 was up-regulated under salt, drought,ABA treatment,indicating that PwVQ1 might involved in various stresses in plants.
