不同品系小鼠精子冷冻及冻融精子体外受精的研究

摘    要:采用小鼠精子冷冻、冻融精子体外受精的方法,进行小鼠保种、引种、生物净化及品系标准化的可行性研究,并初步探究小鼠精子冷冻的影响因素。实验选用同一年龄5种不同品系小鼠分别进行精子冷冻、复苏,并比较各品系冷冻效果及冻融精子的体外受精(IVF)率。结果显示,冷冻后各品系小鼠精子复苏存活率约在15%~50%之间;冻融小鼠精子体外受精结果随遗传背景的不同差异显著,受精率分别为:KM小鼠68.4%、ICR小鼠30.3%、DBA/2J小鼠47.2%、C57BL/6J小鼠6.8%、BALB/cA小鼠25%。冻融精子IVF得到的胚胎可以耐受体外培养。

Abstract:Mice are the most commonly used laboratory animals, owing to the availability of large amounts of information on their genetics and biology. As a result, cryopreservation of mouse spermatozoa has recently became available in the mouse model archiving. We attempted to practise the technique of mouse sperm freezing and in vitro fertilization of frozen- thawed sperm in our laboratory. Sperm from KM,ICR,DBA/2J,mBALB/cA and C57BL/6J mouse were frozen in 18% raffinose/3% skim milk cryoprotective solution. The post- thawed sperm from all strains were evaluated for their viability and fertility by comparing them with their fresh sperm as a control. The progressive motility and fertility of sperm in each strain were affected differently after cryopreservation. The fertilization rates of the same strains oocytes inseminated with frozen- thawed spermatozoa were 68.4% ,30.3% ,47.2% ,25% and 6.8% respectively in KM,ICR,DBA/2J,BALB/cA and C57BL/6J mouse. These results indicate that cryopreservation of spermatozoa provides an effective alternative to embryos freezing and corroborate that successful cryopreservation is related to genetic background of mouse strains.
