摘 要: | 目的]采用pET-28a系统包涵体表达死亡素-蜂毒素杂合肽。方法]将死亡素基因重组到pET-28a质粒上,转化到大肠杆菌BL21中构建基因工程菌,SDS-PAGE电泳检测工程菌诱导表达时间,亲和层析纯化目的蛋白。结果]表达的目的蛋白为包涵体,工程菌诱导表达时间8h最佳。结论]包涵体方式表达死亡素-蜂毒素可以获得较高的表达量,纯化后蛋白纯度较高。

Abstract: | Objective]The research aimed to express thanatin-melittin hybrid peptide with pET-28a expression system.Method]Recombinant plasmid inserted thanatin-melittin gene was transformed into E.coli BL21,and culturing time of engineering bacterial after inducing was studied by SDS-PAGE electrophoretic analysis.Recombinant protein was purified by affinity chromatography.Result] Recombinant protein was in inclusion body,and ideal inducing time was 8 hours.Conclusion]High expression and purification of thanatin-melittin hybrid peptide could be gotten. |
