牛乳头瘤病毒基因2型流行株全基因组的克隆及序列分析

摘    要:为了解牛乳头瘤病毒(Bovine Papillomavirus,BPV)新疆流行株基因型及其基因组遗传变异情况,用乳头瘤病毒L1基因简并引物 FAP59/FAP64对新疆沙湾县奶牛皮肤肿瘤组织样品进行PCR扩增,测序后确定,BPV基因型;设计6对BPV基因型特异性引物,经扩增、测序、拼接后获得BPV流行株全长基因组序列。 L1基因序列分析表明,BPV2 SW01流行株为BPV 2基因型;该毒株基因组全长7 944 bp,包括 E6、E7、E1、E8、E2、E4、E3、E5、L2L1 10个开放阅读框(ORF)。遗传进化树分析显示,BPV2 SW01归于Delta乳头瘤病毒属,与BPV 1和BPV 13进化关系最近。

Abstract:In order to clarify the genotype and better understand variation of genome of the bovine papilloma virus (BPV), DNA extracted from skin tumors of Xinjiang Shawan cow was used as temple to detect PBV infection by PCR using the primer pair of FAP59/FAP64. The amplified fragment was then sequenced, analyzed and genotyped. Based on the genotyping results 6 pairs of specific primers of BPV were designed and used to amplify the genome of BPV epidemic strains. Sequence analysis of L1 gene showed that BPV2 SW01 belonged to the BPV 2 genotype. The genome length was 7 944 bp, including 10 open reading frames (ORFs). Phylogenetic analysis showed that BPV2 SW01 was grouped within the genus Deltapa pillomavirus, which is most closely related to BPV 1 and BPV 13.
