利用圆环病毒2结构蛋白建立检测其抗体

摘    要:为建立检测PCV2抗体的间接酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA),将猪圆环病毒2(porcine circovirus type 2,PCV2)结构蛋白Cap(不含核内化信号肽)的基因克隆到原核表达载体pET28a(+)中,并在大肠杆菌中表达,获得可溶性的重组蛋白(Cap⊿41),用Cap⊿41作为诊断抗原,确立了间接ELISA的最佳反应条件,并对来自4个猪场的394份血清进行检测。结果表明,与免疫荧光检测相比,建立的间接ELISA的检测敏感性和特异性分别为93.14%和95.72%,且建立的间接ELISA的批内与批间差异较小。此外,对394份血清的检测结果体现的4个猪场的PCV2抗体消长规律与临床结果相符,表明建立的间接ELISA方法可靠,可以用于PCV2抗体的检测。

Abstract:The strategic emerging industry fusing emerging technology and emergent industry is not only direction of modern scientific innovation, but also guide industrial development, it will satisfy developmental orientation of ecological economy. Because of the above consideration, the article elaborate on the characteristic of strategic emerging industry from the standpoint of driving function,ecological function and technical function.Through index system''s selection and the utilization of Weaver-Thomas model, the article has established a set of systematic appraisal system based on the characteristic. Aiming at the selection of the regional strategic emerging industry, the last part of the paper made a demonstrative analysis to 8 industrial groups developmental condition of Hunan Province and the result conforming to the really developmental condition indicate that the appraisal system is scientific and rational.
