花生种子相对分子质量17500多肽抗体制备及其应用

摘    要:用一种纯化的花生种子高甲硫氨酸蛋白-相对分子质量17500多肽为抗原,免疫动物获得了高特异性的抗血清,以之为探针分析比较了其它几种豆科植物中们花生球蛋白Ⅱ提取物,免疫亲合反应均呈阳性,而玉米种子呈阴性反应,表明该多肽是豆科植物种子蛋白保守成分,Western blot分析表明该多肽在高甲硫氨酸类型代表品种和低甲硫氨酸类型代表品种中均存在。

Abstract:Antibody from peanut seeds was raised in rabbit. ELISA using the antibody against peanut Mr 17 500 polypeptide for conarachin II in seeds of soybean, mung bean, cowpea and corn was analyzed. Results showed that there were immunological cross reactions of that antibody with the protein components in the soybean, mung bean and cowpea seeds, but no immunological cross reaction with the protein components in corn. Western blot showed that there was Mr 17 500 polypeptide in both peanut cultivars with different methioine composition.
