摘    要:红魔芋是自然界中含有大量葡甘聚糖的经济作物之一,其繁殖系数较低,病毒感染及种质退化严重。为解决红魔芋繁殖系数低、种质退化等问题,以红魔芋的球茎和无性株茅为外植体进行组织培养试验。研究表明,地下球茎是最佳外植体;75%的酒精浸泡30s,再用0.15%的升汞浸泡30min是最理想的消毒方式;愈伤及不定芽最佳培养基为MS+6-BA2mg/L+NAA0.1mg/L。

Comparative Study on the Adventitious Shoots Inducing Different Explants in Vitro in A.xiei

Abstract:A.xiei is one economic crops with rich-konjac glucomannan in nature, but its propagation was lower, and the plant virus infection and germplasm degradation was severe. To solve the problem of low propagation coefficient and germplasm degradation in A. xiei, two exptants: bulbs and bulbils, were used for propagation in vitro in tissue culture in A.xiei. The results show that the underground bulb is the best explants; the best way to disinfect is to soak the explant in 0.15% mercuric chloride for 30minutes after soaking it in 75% alcohol for 30 seconds; the best medium for inducing callus and bud is: MS+6-BA2mg/L+NAA0.1mg/L.
