黄粉虫胰蛋白酶酶解工艺的研究

摘    要:以黄粉虫为原料,以水解度(DH)和水解多肽液的还原能力为指标,研究胰蛋白酶法制备黄粉虫多肽的工艺条件。通过单因素试验,初步确定pH、温度、酶用量、固液比的正交试验水平;在单因素试验基础上设计正交试验,分析各因素对酶解反应的影响及确定酶解反应的最佳工艺条件。结果表明:pH对酶解反应的影响最大,其次是温度和酶用量,固液比对酶解反应的影响最小;酶解的最佳工艺条件为:pH为8.5,温度为46℃,酶用量为1.8%,固液比为1∶10,酶解时间为4h,DH为13.223%。

On Enzymatic Hydrolysis of Protein in Tenebrio Molitor with Trypsin

Abstract:The enzymatic hydrolysis of proteins from Tenebrio Molitor with trypsin was investigated.The degree of hydrolysis(DH) and deoxidization capability were employed for the evaluations of effect or function in the hydrolysis.The single factor experiment gave the levels of the main parameters,including enzyme dosages,ratios of solid to liquid,temperatures and pHs.Based on the results from single factor experiment,the orthogonal experiment was designed and conducted.The results showed that the most factors in protein hydrolysis of Tenebrio Molitor with trypsin was pH,followed by temperature and enzyme dosage,the last factor was ratios of solid to liquid.The optimal conditions of hydrolysis were as follows: pH 8.5,temperature 46℃,enzyme dosage 1.8%,ratios of solid to liquid 1:10,hydrolysis duration 4 h,DH 13.223%.
