海南中华蜜蜂线粒体DNA的遗传多样性

摘    要:采用线粒体DNA tRNAleu-COII片段的序列分析方法,对海南16个样点的715群中华蜜蜂(Apis cerana cerana)进行遗传多样性研究,明确了海南中华蜜蜂线粒体DNA tRNAleu-COII片段序列的遗传多样性及单倍型的分布特点.结果共发现48种单倍型,其中新发现43种单倍型.共检测到38个变异位点,包括12个单一多态位点、23个简约信息位点、1个插入位点和3个缺失位点.海南中华蜜蜂单倍型多样度为0.742±0.017,平均核苷酸差异数为1.248,核苷酸多样度为0.00354±0.00013,这表明海南中华蜜蜂线粒体DNA tRNAleu-COII片段具有丰富遗传多样性.海南中华蜜蜂普遍在117位点发生G→A的转换,发生比例为74.27%.

Abstract:Genetic diversity of 715 colonies of Apis cerana cerana from 16 sampling spots throughout Hainan Province in China were surveyed by mitochondrial DNA fragments of tRNAleu-COII sequence.Total 48 haplotypes were found,43 of which had not been previously published.There were 38 polymorphic sites,including 12 singleton variable sites,23 parsimony informative sites,1 insert site and 3 deletion sites.The average diversity of haplotypes was 0.742±0.017,the average nucleotide difference and the nucleotide diversity of the honeybee samples were 1.248 and 0.00354±0.00013,indicating the abundance of the haplotypes.There were 74.27% of honeybees in Hainan detected the transition in 117 site(G→A).Honeybee introduced from the mainland and artificial rearing queen have led to the unstable stage of the genetic structure of Hainan honeybee in the influenced regions,that was why there was a significant difference genetic structures between sampling sites in the same region.
