川芎中阿魏酸类效应组分的提取纯化工艺研究

摘    要:目的]优化川芎中阿魏酸类效应组分的提取纯化工艺条件。方法]以溶剂量、提取次数、乙醇浓度、提取时间为考察因素,以阿魏酸类组分为检测指标,采用正交试验优化川芎中阿魏酸类效应组分的提取工艺;以树脂类型、上样量、除杂溶剂、洗脱溶剂等为考察因素,以阿魏酸为测定指标,优化阿魏酸类组分的纯化工艺。结果]确定川芎中阿魏酸类组分优化的提取工艺为:料液比1:8(g/m1),乙醇浓度70%,提取3次,每次0.5h;阿魏酸类组分的纯化树脂为HP300大孔吸附树脂,上样量为5:1(川芎:树脂),2Bv水除杂,2BV浓度50%乙醇洗脱,回收溶剂、真空干燥,纯化物中阿魏酸和总酚酸的含量分别为9.71%和18.69%。结论]所优化的提取纯化条件为川芎效应组分的进一步研究开发奠定了理论基础。

Optimization of Extraction and Purification on Ferulic Acid Ingredients from Chuanxiong Rhizoma

Abstract: Objective ] To optimize the extraction and purification of ferulic acid ingredients from Chuanxiong Rhizoma. Method] The effects of solid-liquid ratio, extract times, ethanol concentration and extract time were investigated to obtain the extraction technology of the ferulic acid ingredients from Chuanxiong Rhizoma according to the orthogonal test. The purification of the resins for ethanol extracts investigated the resin type, sample amount, cleaning solvent and elution solvent. Result ] The optimal technical conditions were extracted twice with 70% ethanol on the ratio of 1 g: 8 ml for femlic acid ingredients when treatment time was 30 minutes. The organic acid of Chuanxiong Rhizoma with HPD300 macroporous resin was absorbed effectively. The proportion of the crude drug and drying macroporous resin was 5: 1. The extract was added to the column of macroporous resin, then washed with distilled water and 50% alcohol, respectively. The 50% alcohol eluent was collected, in which the content of ferulic acid and total phenolic acid was 9.71% and 18.69 % in solids, respectively. Conclusion ] The study explores a practical extraction method for ferulic acid ingredients from Chuanxiong Rhizoma and lays a good foundation for its further research.
