东方伊萨酵母菌对偶氮染料酸性大红GR的脱色优化及特性研究

摘    要:目的]优化东方伊萨酵母菌(Issatchenkia orientalis YP-1)对偶氮染料酸性大红GR的脱色条件,初步阐明脱色机理。方法]研究该菌在不同温度、pH、接种量、染料起始浓度、碳源和氮源条件对酸性大红GR的脱色影响,并且对脱色降解功能酶进行定位。结果]该菌在16 h内对浓度为50~100 mg/L酸性大红GR的脱色率达96%,其机理主要为降解脱色。东方伊萨酵母菌YP-1的最佳接种量不应低于10%,最佳pH在4~8之间,最佳温度为28℃,(NH4)2SO4浓度在0.1%以上,葡萄糖浓度不应低于1%。脱色后染料的主体结构被破坏,发生降解反应。该菌主要脱色降解功能酶为胞内酶。结论]东方伊萨酵母菌对酸性大红GR的脱色效果显著,在染料废水处理中将具有良好的应用前景。

The Decolorization Optimization and Characteristics of an Azo Dye Acid Scarlet GR by Yeast Issatchenkia orientalis YP-1

Abstract: Objective ] The study aimed to discuss the optimal conditions and preliminary decolorization mechanism for the decolorization and degradation of azo dye Acid Scarlet GR by yeast strain Issatchenkia orientalis YP-1. Method] The effect of different temperatures, pH, inoculation volumes, initial dye concentrations, carbon and nitrogen concentrations on decolorization with I. orientalis YP-1 was discussed in detail, as well as its preliminary decolorization mechanism. Result] This strain achieved a maximal decolorization (96%) for Acid Scarlet GR (50 -100 mg/L) after incubation for 16 h. To get such decolorization, the optimal inoculation volume of this strain was set not less than 10%, optimal pH of culture medium ranging from 4 to 9, and concentrations of ( NH4 )2SO4 and glucose not lower than 0.1% and 1% , respectively. In addition, the results showed that the main structure of acid scarlet GR was destroyed after decolorization and the main functional decolorization enzyme was intracellular enzyme. Conclusion] The yeast strain lssatchenkia orientalis YP-1 has relatively strong decolorization capability on azo dye acid scarlet GR, which has a good prospect in the biodegradation of dye wastewater.
