京郊设施蔬菜黄瓜花叶病毒的检测

摘    要:为了明确北京地区设施蔬菜上常见病毒的发生情况,利用ELISA和RT-PCR方法对京郊设施蔬菜中的黄瓜花叶病毒(Cucumber mosaic virus,CMV)进行检测。共采集设施栽培的芹菜、观赏南瓜、结球生菜、大南瓜、辣椒、抱子甘蓝、番茄(‘大红袍’和‘红罗马’品种)、四棱豆、茄子共10个蔬菜作物样品的叶片进行测定。结果显示:芹菜、观赏南瓜、结球生菜、辣椒、茄子和番茄‘大红袍’品种的样品含有CMV,大南瓜、抱子甘蓝、四棱豆和番茄‘红罗马’品种的样品未检测到CMV。ELISA和RT-PCR检测结果一致,将检测结果为阳性样品的RT-PCR产物进行了序列测定和分析,验证了检测结果的可靠性。

Abstract:Cucumber mosaic virus (CMV) was detected by ELISA and RT-PCR methods to investigate the distribution and diversity of viruses infecting vegetable crops grown in Beijing suburban greenhouses. Leave samples of 10 vegetable crops were collected and then tested. Identical results were obtained by both ELISA and RT-PCR methods showed that 6 samples of celery, ornamental pumpkin, lettuce, peppers, eggplant and tomato ‘Dahongpao’ were positive while the other 4 samples of great pumpkin, brussels sprouts, four winged bean and tomato ‘Rome red’ were negative to CMV. In our study, the reliability of the results was confirmed by sequencing and sequence analysis of the RT-PCR products from the positive samples.
