| 摘 要： | 选用体细胞胚高频发生的黑龙江省主栽大豆品种 (东农 7819)的未成熟子叶 ,在含高浓度的生长素 MSB培养基上诱导体细胞胚的产生。体细胞胚分别置于 M3、M10、MC三种成熟培养基上或单纯置于灭菌过的培养皿进行干化处理。成熟处理为大豆体细胞胚萌发所必需。单纯干化处理不利于体细胞胚存活 ,而在高渗成熟培养基 MC或 M10成熟处理过的体细胞胚在 MSG(1/2 MSB+ GA)萌发培养基上的萌发率明显高于 M3。成熟处理后 ,形态正常体细胞胚的萌发率达90 % ,明显高于形态不正常胚 4 %的萌发率。对成熟处理过的体细胞胚的组织学研究表明 ,形态学上苗端未分化是形态不正常胚不能形成完整植株的原因之一。

| Abstract: | Soybean somatic embryos were induced from cultured Dongnong 7819 immature cotyledons in the presence of a high level concentration of Naphthakenacetic acid(NAA).Maturation culture was necessary for somatic embryos germanation.Maturation conditions of somatic embryos were optimizing.Indirect desiccation of somatic embryos on the high osmotic mediums (Mc or M10) were better than the direct desiccation in petri dish.Regeneration frequncy of normal embryos were 91%,higher than abnomal embryos via maturation culture.Histological studies showed:apical meristems undifferentiation of some abnomal embryos was one of factors which affect production of complete plant. |
