香青兰的离体保存研究

摘    要:以香青兰为试材,研究不同养分水平培养基,不同浓度甘露醇、多效唑及琼脂对试管苗离体保存的影响.结果表明:1/4MS培养基中试管苗前期存活率较低,但保存时间延长了4个月以上:甘露醇对试管苗的生长有抑制作用,可延长试管苗存活时间4个月;多效唑能促进香青兰试管苗侧芽分化,但对试管苗茎的伸长有明显抑制作用,其中浓度为2.0 mg/L时试管苗存活率最高,保存时间可延长4个月,保存至9个月时,存活率达到50%;MS培养基中附加琼脂浓度8.5~10.5 g/L为香青兰最佳离体保存方法,该条件下香青兰试管苗保存时间明显延长,保存12个月时存活率仍高达85 %~60%,且试管苗恢复生长后长势良好,其再生后代的形态特征与对照株无差异.

Abstract:The effect of different concentrations of mannitol, paclobutrazol and agar on Dracocephalum moldavica L. in vitro plantlet preservation were studied. The results showed that the survival rate of the plantlets growing in the condition of 1/4MS medium was lower than that in the condition of MS and 1/2MS medium at earlier stage, however its survival time had extended for more than 4 months. The growth of vitro plantlet was significantly inhibited by using mannitol, however its survival time had also extended for more than 4 months. The growth of vitro plantlet was significantly promoted by paclobutrazol, the survival rate of vitro plantlet was 50% in the condition of 2.0 mg/L which could extend for 4~9 months. MS medium added with agar 8.5~10.5 g/L was the best condition for the growth of Dracocephalum moldavica L. in vitro plantlet, which couod be saved for 12 months with a high survival rate of 85%~60%, and the plantlets grew well with no difference compared with the control groups.
