COX-2,MMP-9和bFGF在大肠癌中的表达及临床意义

摘    要:目的探讨COX-2,MMP-9和bFGF在大肠癌发生、发展及浸润转移中的作用及相互关系.方法应用免疫组化染色(SP)法检测COX-2,MMP-9和bFGF在66例大肠癌及22例正常大肠组织的表达情况.结果大肠癌组织COX-2,MMP-9和bFGF阳性表达率分别为84.8%,75.8%,65.2%,与正常大肠组织的27.3%,9.1%和4.5%相比有显著性差异(P〈0.01),COX-2表达与肿瘤组织的浸润深度和淋巴结转移显著相关(P〈0.05);MMP-9表达与淋巴结转移显著相关(P〈0.05);bFGF表达与肿瘤组织的浸润深度显著相关(P〈0.05);Spearman等级相关检验显示,COX-2的表达与MMP-9和bFGF的表达显著相关(P〈0.05,P〈0.01).结论COX-2,MMP-9和bFGF在大肠癌的发生、发展中发挥着重要作用,COX-2可能通过促进MMP-9和bFGF的表达,从而促进大肠癌的浸润和转移.

Expression and Clinical Significance of COX-2, MMP-9 and bFGF in Colorectal Carcinoma

Abstract:Objective To investigate the expression of COX-2,MMP-9 and bFGF in colorectal carcinoma tissue and their interactions with clinical pathological characteristics. Method Sixty-six patients with colorectal carcinoma were obtained, and twenty-two corresponding paracancerous normal tissues were randomly selected as control. Immunohistochemical staining was used for detecting the expression of COX-2, MMP-9 and bFGF. Results The positive expression rates of COX-2, MMP-9 and bFGF in colorectal carcinoma were 84. 8% , 75.8% ,65.2% respectively, which were significantly higher than those in the normal tissue (P 〈 0.01 ). The expression of COX-2 has a significant relation with invasion depth of tumor, lymphnode metastasis (P 〈 0.05 ) ; The expression of MMP-9 has a significant relation with lymphnode metastasis (P 〈 0.05 ) ; The expression of bFCF has a significant relation with invasion depth of tumor ( P 〈 0. 05 ). In addition, Spearman rank correlation test showed that the expression of COX-2 and MMP-9, bFGF were significantly related with each other (P 〈 0.05 ). Conclusion COX-2, MMP-9 and bFGF play very important roles in the development of colorectal carcinoma. COX-2 contributes to development of colorectal carcinoma by promoting MMP-9 and bFGF expression.
