禽淋巴细胞性白血病的诊断与病毒亚群鉴定

摘    要:采用病理组织学方法对某商品蛋鸡场送检的发病鸡进行实验室诊断,在肿胀的肝、脾、肾和法氏囊组织切片中观察到成淋巴细胞增生病变。使用禽白血病A、B亚群抗体检测试剂盒对血清样本进行间接ELISA试验,抗体阳性率达到44.4%(12/27)。采集12只病死鸡的肝脏材料提取DNA样本,11份样本中扩增出ALV-A亚群特异性的病原核酸(691bp),检出率达到91.6%(11/12)。将扩增的目的基因克隆与测序,截取gp85基因片段可变区序列与ALV-A、B、C、D和E亚群参考毒株的相应序列进行比较,同源性分别达到89.0-89.6%、67.1-67.7%、69.1-70.1%、69.1-69.5%和70.9-72.0%。结果证实本次疫病是由ALV-A亚群病毒引起的淋巴细胞性白血病。

Abstract:Histopathological method was used for the diagnosis the diseased chickens in a commercial layer farm. The lymphoblast proliferative lesions were observed in tissue slides of swollen liver,spleen, kidney and Fabricius bursa.The serum samples were detected using avian leukosis virus antibody test kit-subgroup A,B (ALV-AB) and the antibody positive rate reached 44.4%(12/27). The total DNA samples were extracted from liver materials of 12 dead chickens. A 691 bp fragment was amplified from 11 DNA samples,which was specific for the subgroup A of ALV and the detective rate accounted for 91.6% (11/12).The amplified target genes were cloned and sequenced. The comparison was made against the variable region of gp85 gene with subgroup A、B、C、D and E of ALV. The homology reached 89.0~89.6% 、67.1~67.7% 、69.1~70.1% 、69.1~69.5% 和 70.9~72.0% respectively. It was demonstrated that the lymploid leucosis was caused by subgroup A of ALV.
