土壤放线菌分离方法研究

摘    要:研究了土壤放线菌的分离方法,结果表明:培养基中加入150mg/L重铬酸钾,既有利于放线菌的生长,又能抑制真菌和细菌的生长;土样在28℃或室温风干10~20d后,制成10%土壤悬浮液,40~50℃、pH7条件下,加6%酵母膏(或6%蛋白胨)和0.05%SD(S5mmol/L磷酸缓冲液),振荡10~20min,灭菌水稀释成1%后,与含250mg/L重铬酸钾的高氏1号琼脂培养基制成平板,最有利于放线菌的分离。

Abstract:The optimum concentration of the inhibitor potassium dichromate(K2Cr2O7) in the plate medium was studied;meanwhile,the study on pretreatment of soil samples collected from corn field was conducted.The results showed that the suitable method of the selective isolation of actinomycetes was: after the collected soil suspension(10 %) added with 6 % yeast extract and 0.05 % SDS(5 mmol/L phosphate buffer)was stirred at 40 ℃,pH 7 for 20 minutes,or the soil suspension(10 %) added with 6 % peptone and 0.05 % SDS(5 mmol/L phosphate buffer) was stirred at 50 ℃,pH 7 for 10 minutes,then the soil suspension diluted with water to 1 % was inoculated with the method of plate dilution on Gause's starch agar added with 150 mg/L K2Cr2O7 medium for 7 days and these pretreatments were effective on actinomycetes' isolation.
