摘    要:目的]在对OsWNK基因的生化特性有了一定了解的基础上,研究OsWNK在水稻根组织的表达,为进一步研究基因OsWNK的功能奠定基础。方法]利用RNA原位杂交技术,通过制备OsWNK基因的正反义地高辛标记的RNA探针,与水稻根内靶基因转录表达产物进行原位杂交定位。结果]在根尖生长点部位的OsWNK表达最旺盛,而在伸长区的细胞中,OsWNK的表达量很低。另外,在根冠中几乎没有OsWNK的表达。结论]OSWNK在根尖生长点的大量分布说明仍聊忧在植物快速生长的组织和细胞中表达比较高,推测此基因可能与生长点细胞频繁的细胞分裂有关。

Research on the Expression Pattern of OsWNK in Rice Root Tissue

Abstract:Objective] On the basis of understanding of the biochemical performance of OsWNK,its expression pattern in rice root tissue was explored to lay the foundation for the research on OsWNK function.Method] With in situ RNA hybridization technique,the sense and anti-sense OsWNK probes were hybridized with the products expressed by the target gene in rice root tissue.Results] The experimental results showed that OsWNK had a high expression in the growing point cells of root tip,while its expression was very weak in elongate area cells and there was no expression detected in root cap cells.Conclusion] OsWNK was mainly accumulated in the fast growth cells and tissues such as growing point cells of root tip,which meant that OsWNK might play roles in cell division.
