| 摘 要： | 通过试验比较，分析了CTAB法中EDTA（乙二胺四乙酸）浓度和PVP（乙稀吡咯烷酮）对碗莲叶片DNA提纯结果的影响；同时还比较了叶片采集时间及遮光处理对提纯结果的影响。结果确定了适于碗莲叶片基因组DNA提取的方法以及如何选择最合适的材料进行DNA的提取，从而获得RAPD、RFLP、SSR等分子标记手段所需要的高纯度基因组DNA。

| Abstract: | The influences of EDTA concentration, PVP, sampling time and shading treatment on the DNAs extraction from Nelumbo nucifera leaves were studied. The appropriate method for the DNAs extraction from Nelumbo nucifera leaves and most suitable leaves for sampling were determined. The very pure extracted DNAs could be used directly for RAPD,PFLP,SSR analysis which are useful for molecular markers. |
