裂拟迷孔菌生物学特性及驯化栽培分析

摘    要:为了发掘与利用多孔菌资源,对采自辽宁省白狼山国家级自然保护区白腐真菌裂拟迷孔菌(Daedaleopsis confragosa)分离纯化得到的菌株,从碳源、氮源、pH值、温度4个方面进行生物学特性和驯化栽培试验研究,从中选出3个较优水平进行正交试验。结果表明,单因素试验中最适碳源是可溶性淀粉,最适氮源为磷酸氢二铵,最适初始pH值为8,最适生长温度为30℃。最适培养条件下对裂拟迷孔菌液体发酵菌种进行锰过氧化物酶(manganese peroxidase, MnP)活力测定,得出酶活在第8天到达峰值19.411 U·L-1,具有一定锰过氧化物酶活性。出菇试验中,二级种选用麦粒菌种,25℃培养。裂拟迷孔菌可在人工栽培下形成子实体,培养料配方为阔叶树木屑78%、麸皮20%、石膏1%、蔗糖1%,含水量65%,从接种至子实体成熟需要54 d。

Abstract:In order to explore and utilize the resources of polypores, the biological characteristics and domestication cultivation of the strains isolated from the white rot fungus Daedaleopsis confragosa collected from Bailangshan Mountain National Nature Reserve in Liaoning province were studied from 4 aspects: carbon source, nitrogen source, pH and temperature. The optimal carbon source was soluble starch, and the optimal nitrogen source was diammonium hydrogen phosphate. The optimum pH is 8.The optimum temperature is 30℃. The activity of manganese peroxidase(MnP) in D. confragosa liquid fermented seeds was determined under optimal culture conditions, and the enzyme activity reached the peak of 19.411 U·L-1 on the 8 th day with manganese peroxidase activity. The spawn was selected from the wheat grain strain and cultured at 25°C. D. confragosa could form fruiting bodies under artificial cultivation. The culture formula is 78% broadleaf wood chips, 20% bran, 1% gypsum, 1% sucrose and 65% of water content, and it takes 54 days from inoculation to fruiting body maturation.
