微型月季组织培养快繁体系的建立

摘    要:以微型月季品种“旋转木马(红色系)”和“金太阳(黄色系)”为试验材料,研究组织培养快繁体系适宜的外植体,外植体萌发适宜的培养基;丛生芽增殖适宜的培养基;组培苗生根的最佳培养基及移栽条件,以期建立微型月季快繁体系,提高种苗繁殖速度和质量。结果表明:微型月季适宜组织培养的外植体为1年生嫩枝和半木质化茎段;适宜外植体腋芽萌发的培养基为MS+0.5mg/L6-BA;适宜的增殖培养基为MS+0.5mg/L6-BA+0.5mg/LTDZ或者MS+1mg/LTDZ+0.1mg/LKT;适宜的生根培养基为0.2mg/LIBA;组培苗根系长至0.5~1cm、株高2~3cm时进行移栽,移栽后遮光率20%~40%、空气湿度40%~50%、环境温度20~25℃,移栽成活率可达到91%。

Abstract:In this experiment, the materials is miniature rose of Merry-go-round(Red) and The golden sun (Yellow),we study suitable for growth of explant and medium;Plant growth regulator effects on adventitious bud proliferation;The best culture medium and transplanting conditions for tissue culture seedlings, in order to establish miniature rose tissue culture rapid propagation system. It will be used to improve the speed and quality breeding. The results show that the best of explants is annual shoots of twigs and semi-lignified stem segments. optimum adventitious bud regeneration medium for MS + 0.5mg/L 6-BA;Best adventitious bud proliferation culture medium for MS+0.5mg/L 6-BA+0.5mg/L TDZ or MS+1mg/L TDZ+0.1 mg/L KT;The best rooting culture medium for 0.2 mg/L IBA;We transplant tissue culture seedling,when it grew to 0.5-1 cm of roots and 2-3 cm tall of plants;After transplanting Shading coefficient is 20%-40%, humidity of the air is 40%-50%, environment temperature is 20-25 ℃, transplanting survival rate can reach 91%.
