八个猪种PRLR和RBP4基因PCR-RFLP检测及群体遗传特性研究

摘    要:本研究采用PCR-RFLP方法,对莱芜黑猪、鲁莱黑猪、里岔黑猪、鲁烟白猪、新沂蒙黑猪5个山东本地猪种和大约克夏、长白、杜洛克3个引进猪种共八个猪种323头繁殖母猪进行PRLR和RBP4基因的多态性检测,并对其遗传多样性进行分析。结果表明:两个基因位点在八个猪种的测定群体中均存在多态性,在三种基因型中AB基因型出现频率最高,山东本地猪种与引进猪种间基因型频率存在显著差异(P<0.05)。八个猪种PRLR和RBP4基因位点的期望杂合度分别在0.3265~0.5062和0.2655~0.5019之间,多态信息含量分别在0.2688~0.4268和0.2264~0.3736之间。研究结果提示:杜洛克猪在这两个基因位点的杂合度最低,其遗传一致性最好,其余猪种均表现为中度多态,表明这些猪种具有较为丰富的遗传多态性。

Abstract:323 sows,including five Shandong local pig breeds and three foreign pig breeds,were collected to detect the polymorphisms of prolactin receptor(PRLR) and retinol-binding protein 4(RBP4) genes by PCR-RFLP method,and the genetic diversities were analyzed.The results showed that the polymorphic sites of both PRLR and RBP4 genes were found in all populations tested,and the genotype frequency of AB was higher than that of AA and BB.The genotypes distribution revealed great differences between Shandong local pig breeds and foreign pig breeds(P<0.05).For PRLR and RBP4 genes,the expectative heterozygosity were 0.3265~0.5062 and 0.2655~0.5019,the polymorphism information content is 0.2688~ 0.4268 and 0.3419~0.3736,respectively.It implied that the Duroc pigs had highest homogeneity.The other pig breeds,however,had abundant genetic diversity.
