高产乳酸链球菌素生产菌的诱变

摘    要:为了获得高效价的Nisin生产菌株,采用传统的微生物育种技术,以Nisin生产菌乳酸乳球菌作为出发株,通过紫外照射和亚硝酸处理进行协同诱变育种。根据致死率和诱变剂量的相互关系,分别得出了紫外线照射和亚硝酸诱变的致死率曲线以选择合适的诱变剂量。最终确定采用2.5 min紫外线照射和4 min亚硝酸处理对乳酸乳球菌进行诱变获得突变菌株。突变株经初筛和复筛得到Nisin高产菌株,其发酵液Nisin的效价较出发株高1.72倍。

Abstract:The traditional microbiological breeding technology was used to obtain a high Nisin-producing Lactococcus lactisstrain.The parental Nisin-producing strain was treated with UV and nitrous acid.According to the correlation of lethal rate and inducing dose,the lethality curve of UV and nitrous acid treatment were obtained in L.lactis,and the optimum treatment dose was determined.The mutant strain was obtained by induced breeding of parental strain after being treated with 2.5 min UV and 4 min nitrous acid.Afte...
