秋水仙素诱导野生杜鹃花变异试验

摘    要:应用0.5、1.0、2.0、3.0 mg/mL 4种浓度的秋水仙素溶液诱导正在发育的映山红(Rhododendron simsii)、马银花(Rh.ovatum)、满山红(Rh.mariesii)的花芽和叶芽,观察其花粉粒及气孔变异情况.结果表明,3种杜鹃叶芽和花芽表现出的多倍性有一定的变化,映山红花粉粒在3.0 mg/mL的浓度处理时变异最大,增加37.99%;满山红花粉粒在2.0 mg/mL的浓度处理时变异最大,增加21.96%;随着秋水仙素浓度的增加,3种杜鹃的气孔器大小变异增加,在3.0 mg/mL浓度处理时增加最多,出现气孔增长、保卫细胞宽增加、保卫细胞长度增加等多倍体表现;气孔密度变化与多倍性联系规律不明显.

Abstract:Induction was conducted on developing flower and leaf bud of Rhododendron simsii, R. ovatum and K mariesii with four treatments of colchicines concentrations, 0.5mg/mL, 1.0 mg/mL, 2.0 mg/mL and 3.0 mg/mL. Observation on microspore and stomata of tested leaf and flower buds demonstrated polyploid variation. Microspore of K simsii increased 37.99% with 3.0 mg/mL of colchicines while that of R. mariesii increased 21.96% with 2.0 mg/mL. Stomata of three tested Rhododendrons varied with increase of colchicines concentration topped at 3.0 mg/mL. The variation of stomata density had no evident relation with polyploid.
