海岛棉枯萎病抗性遗传规律及分子标记的初步筛选

摘    要:目的]研究海岛棉枯萎病抗性遗传规律,并对与枯萎病抗性基因连锁的分子标记进行初步筛选.方法]以高产、优质、高感枯萎病的海岛棉品种新海21号为母本、高抗枯萎病的海岛棉品系HK237为父本,构建了P_1、P_2、F_1、F_2、F_(2:3)家系、BC_1六个世代群体.在苗期于温室中用人工接菌法对各个世代群体的枯萎病抗性鉴定.结果]采用BSA法对2 000对SSR引物进行筛选,共获得5对在亲本及抗、感DNA池表现多态性的引物.根据多态性标记对F_2群体基因型鉴定结果结合抗病性表现,利用Mapmaker/Exp(Version3.0b)统计分析软件进行遗传连锁分析.结果表明,标记NAU3240与海岛棉枯萎病抗性位点之间紧密连锁.结论]海岛棉HK237对枯萎病的抗性遗传受显性单基因控制,表现为质量性状的遗传规律.

Inheritance of Resistance to Fusarium wilt and their Molecular Marker in Gossypium barbadense
Abstract:【Objective】This experiment planed to study the inheritance of resistance to Fusarium wlit and screen their molecular marker in Gossypium barbadense.【Method】This study took the susceptible G.barbadense Xinhai21 and the resistant G.barbadense HK237 as parents,build P1,P2,F1,F2,F2:3 and BC1 separate groups.The resistance of the F2∶3 was determined in the greenhouse.All of the F2∶3 lines were tested to verify the resistance of the relevant F2 individuals.【Result】Two thousand SSR primers were screened using the BSA method.Out of these primers,five pairs of primer showed polymorphic between parents and resistant,susceptible DNA pools.It was found that NAU3240 linked with the gene of Fusarium wilt resistance when calculated by Mapmaker/Exp(Version3.0b) software.【Conclusion】According to the result,we found that the resistance to Fusarium wlit of HK237 was qualitatively inherited trait and controlled by a single dominant gene.
